Sod knockout Sod knockout blunts the lifespan extension by a high sugar-low protein diet, but not a low-calorie diet [22672579]. Fly
Sod2 RNAi RNA interference of Sod2 results in increased oxidative stress and early-onset mortality in young adults [12456885]. Fly
High sugar low protein diet A high sugar low protein diet increases the lifespan, but not resistance to acute oxidative stress [22672579]. Fly
Pinitol supplementation Pinitol (a 3-methoxy analogue of D-chiro-inositol) supplementation to the diet. For both males and females, a 20 microMolar dose of pinitol significantly extends median lifespan by 13% (p < 0.05) and 12.5% (p < 0.05), respectively. Lifespan extension by pinitol is accompanied by protection against oxidative and starvation stresses, improvement in health span, and no reduction in fecundity. Pinitol increases organismal lifespan of both in dietary restriction and ad libitum conditions. Nuclear localization of foxo increases in pinitol-fed animals. Pinitol treatment significantly activates JNK and S6K, but not AKT [22843669]. Fly +12.5 to +13
Low calorie diet with low-sugar content A diet with low-calorie and low-sugar content increase the lifespan, but not resistance to acute oxiditive stress [22672579] Fly
D-chiro-inositol supplementation D-chiro-inositol supplementation to the diet extends adult longevity in both male and female animals. 20 microMolar dose of D-chiro-inositol extends median lifespan by 16.7 (p < 0.001) for males and 13% (p < 0.001) for females. Lifespan extension by D-chrio-inositol is accompanied by protection against oxidative and starvation stresses, improvement in health span, and not reduction in fecundity. Nuclear localization of foxo increases in D-chiro-inositol-fed animals [22843669]. Fly +13 to +16.7
Lazarillo supplementation Extracellular forms of Laz have autocrine and paracrine protecting effects for oxidative stress-challanged Drosophila S2 cells. Local effects of GPI-linked Laz inside and outside the nervous system promote survival upon different stress forms, and extend lifespan and healthspan of the flies in a cell-type dependent manner. Ectopic enhancement of Laz expression increases mean, median, and maximum lifespan. Laz overexpression (via the use of a ubiquitous da-GAL4 driver) increases median lifepan by 28.3% (p < 0.0005). Overexpression of Laz specifically in muscles and brain (via GAL4109(2)80 driver) increases median lifespan by 43.5%. Laz overxpression in dopaminergic and serotenergic neurons and epidermis increases median lifespan bt 31.4% (p < 0.0005) [22846641]. Fly +28.3 to +43.5
Resveratrol supplementation Supplementation with resveratrol extends the lifespan [15254550], but not in always [17875315]. Fly
Oligomycin treatment Oligomycin (a specific inhibitor of complex V) feeding exends lifespan on ad libitum and prevents an increase in longevity under DR (started in the adulthood) in males [19968629]. Fly
Ilp3 mutation Ilp3 null mutants have a normal lifespan under AL and a normal DR response. Ilp2 Ilp3 Ilp5 triple null mutants fail to have a normal response to DR. Their response is right shifted, with mutants being shorter-lived compared to wild-type on low but longer-lived on high yeast concentrations [20195512]. Fly
Ilp5 mutation Abundance of Ilp5 mRNA is reduced under DR. Ilp5 null mutants have a normal lifespan under AL and a normal DR response. Ilp2 Ilp3 Ilp5 triple null mutants fail to have a normal response to DR. Their response is right shifted, with mutants being shorter-lived compared to wild-type on low but longer-lived on high yeast concentrations [20195512]. Fly
Cbs RNAi Cbs RNAi partially abrogates increased lifespan by DR, but has no effect on fully fed animals. Cbs upregulation is required for increased lifespan under low-nutrient conditions. Response of male flies to DR is muted in comparison with females [21930912]. Fly
p53 mutation Globally lack of p53 decreases lifespan [16303568]. Loss of p53 activity slightly shortens the lifespan. Mutants that lack p53 survive well up to 50 days, but mortality rate increases relative to wild-type at later ages. p53 mutant animals are extremely sensitive to irradiation [12935877]. Fly
p53 overexpression Overexpression of wild-type Dmp53 during adult life has no significant effect on lifespan [16303568]. Fly
NF1 mutation NF1 mutants have a shortened lifespan and exhibited increased vulnerability to heat and oxidative stress as well as reduced mitochondrial respiration and elevated ROS production [17369827]. Fly
dnc mutation cAMP phosphodiesterase-deficient dunce mutants have an extended maximum lifespan by about 70% [17369827]. Fly +70
Pi3K92E mutation Heterozyogous mutation in Pi3K92E fails to extend lifespan [11292874] and it is recessive lethal. Fly
Scgdelta deletion Deletion of Scgdelta has detrimental effects on the flight muscles of adult animals and heart function. Median lifespan is reduced 15-30% [17855453]. Fly -15 to -30
rut mutation Two rutabaga mutants, rut1 and rut2080, have significantly shortened lifespans [17369827]. Fly
Trx-2 mutation Trx-2 mutants have a 25% reduction in maximum lifespan and exhibit lower tolerance to oxidative stress while animals carrying multiple copies of Trx-2 exhibit higher tolerance [17567437]. Fly -25
Igf1 overexpression Cardiac specific overexpression of Igf1 results in a 23% increase in median lifespan, though no increase in maximum lifespan [17973971]. Fly +23
dj-1beta mutation Loss of function mutation in dj-1beta shortens maximum lifespan by 40% and results in increased sensitivity to oxidative stress and motor impairments [17651920]. Fly -40
Mt2 overexpression Overexpression of Mt2 extends mean and maximum lifespan [15533947]. Fly
sm overexpression Overexpression of sm in males increases mean and maximum lifespan by 29% and and 16%, respectively [22366109]. Fly
DJ-1alpha RNAi RNA interference of DJ-1alpha shortens maximum lifespan by 13% and results in increased sensitivity to oxidative stress and motor impairments [17651920]. Fly -13
