Factors

We need to know every factor which determines lifespan.

Lifespan factors often but not always originate from defined genetic elements. They are not just genes, by definition they can be anything for which a Classifications schema can be build for that is related to the regulation of lifespan, such entities may include Single-Nucleotide Polymorphism, transcript variants, proteins and their complexes, compounds (i.e. small molecules like metabolites and drugs), etc. A factor should be based on a defined molecular entity or genomic position and been classified. It shall be highly flexible and scalable Concept.

While individual lifespan factors within each species or precise defined molecular entities will be captured within the Lifespan App, Data Entries of the Data App may summarize for instance the relevance of each factor class (e.g. homologous group; chemical derivate of related structure and properties, etc.) as well as draw overall conclusions. o

YKU70 Subunit of the telomeric Ku complex (Yku70p-Yku80p) Deletion fo YKU70 shortens lifespan, but does not accelerate the normal aging process [10521401]. YKU70 null mutants are defective for non-homologous end-joining [8754818] and for telomeric silencing [9635192]. Budding yeast
YKL069W Deletion of YKL069W increases sensitivity to oxidative stress and decreases replicative lifespan [19049972]. Budding yeast
YIA6 Deletion of YIA6 decreases replicative lifespan by 30% in the a strain [18340043]. Budding yeast
YHC3 Yeast Homolog of human Cln3 YHC3 deletion decreases 10-20% shortened lifespan [16435200]. Budding yeast
YHB4 Yeast HemogloBin-like protein 4 sfa1;yhb1 double mutant cancels out the ability of moderate DR to extend replicative lifespan, but not chronological lifespan. Indicating that NO homeostasis during DR-induced replicative lifespan extension is crucial. Deleting YHB1 partially abolished DR-induced replicative lifespan extension, whereas deleting SFA1 alone had no effect. Yhb1 and Sfa1 may play redundant roles [21584246]. Budding yeast
YGR130C Deletion of YGR130C increases replicative lifespan increased by 30% in the alpha strain and increased by 20% in the a strain [18340043], as well as mean and maximum chronological lifespan by 18 and 50%, respectively [22216232] Budding yeast

YFH1 Yeast Frataxin Homolog 1 Deletion of YFH1 decreases replicative lifespan by 50% [15130665]. Budding yeast
YER187W Deletion of YER187W increases replicative lifespan by 30% in the alpha strain [19030232]. Budding yeast

YDR124W Deletion of YDR124W increases replicative lifespan by 30% in the alpha and decreases lifespan by 10% in a strain [19030232]. Budding yeast
YDR089W Deletion of YDR089W decreases replicative lifespan by 30% in the alpha strain [19030232]. Budding yeast
YDL180W YDL180W deletion impairs DR-mediated replicative lifespan extension, but does not change lifespan on AL significantly [22912585]. Budding yeast
