Ensembl gene ID: FBgn0015806
Species: Fly (Taxid: 7227)

Functional description:
Part of the TOR pathway involved in cell growth and size; RPS6-p70-protein kinase; RPS6-p70-protein kinase [Source:FlyBase gene name;Acc:FBgn0015806] [Ensembl]; The gene RPS6-p70-protein kinase is referred to in FlyBase by the symbol Dmel\S6k (CG10539, FBgn0015806). It is a protein_coding_gene from Drosophila melanogaster. There is experimental evidence that it has the molecular function: ribosomal protein S6 kinase activity. There is experimental evidence for 13 unique biological process terms, many of which group under: cellular component organization or biogenesis; biological regulation; response to stress; growth; neuron differentiation; neuron projection development; response to DNA damage stimulus; larval feeding behavior; determination of adult lifespan; multicellular organism reproduction. 26 alleles are reported. The phenotypes of these alleles are annotated with: adult; eye; salivary gland; wing disc; trichogen cell; oocyte; wing blade; wing; ommatidium. It has one annotated transcript and one annotated polypeptide. Protein features are: AGC-kinase, C-terminal; Protein kinase, ATP binding site; Protein kinase, C-terminal; Protein kinase, catalytic domain; Protein kinase-like domain; Ribosomal protein S6 kinase; Serine/threonine- / dual-specificity protein kinase, catalytic domain; Serine/threonine-protein kinase, active site; Serine/threonine-protein kinase-like domain. Summary of modENCODE Temporal Expression Profile: Temporal profile ranges from a peak of high expression to a trough of moderate expression. Peak expression observed within 06-24 hour embryonic stages. [FlyBase]

Observation:

Ubiquitous overexpression of a dominant-negative form of S6k (alias dS6K) increases mean lifespan by 22% and overexpression of a constitutively active form of S6k decreases mean lifespan by 34% at 29°C. Overexpression of a dominant-negative form of S6k protects mutants from deleterious effects of rich food, as if mimicking the effect of DR [15186745].

Interventions:
  • Dominant-negative S6k +22%
  • Constitutive active S6k overexpression -34%

  • Assays: Organismal Lifespan
    Mean: +22 to +34

    Classification:
  • Gerontogene
  • Negative Gerontogene
  • Positive Gerontogene
  • DR-Essential
